When you say using, do you mean SAVING or LOADING? Because you can load a frame without any number or padding and if the Load Single Frame Only checkbox is checked in the PRT Loader, it will be shown on all frames.
You cannot SAVE a single frame without a frame number though because we assume that even if you are saving a single frame, it might be part of a sequence and you just want to overwrite it…
Or did I misunderstand the wish?
Loading. Lemme check that again. I was having issues with that, and assumed that padding was required as when I manually renamed the files with 0000, everything was cool.

If I check “Load Single Frame Only” first, then it’s fine with it. So I was just getting messed up with the order of clicks needed.
When Load Single Frame Only is not checked, PRT Loader takes the current time (based on current scene time, offset, eventual Graph and Range limits) and replaces / adds the frame number for each entry on the list. If the file has already a frame number, it gets replaced. If it does not have one, it gets added. Then you might get an error if that file was not found.
When you check “Load Single Frame Only”, the PRT Loader stops messing with the frame number, assuming that any file name on the list should be taken As Is on every frame, without any modifications. So the number plays no role and you can have it or not - if the verbatim file name exists on disk, it will be loaded.
